    Painting a stripe

    Do you guys think it'd be easier to lay down the stripe color first, then tape it off and lay down the rest, or vice versa?

If it's a color that requires several coats to cover (metallics especially) then do that one first. If it's all solid colors then it really doesn't matter (ie: A GS1000S) Main thing is spraying the least amount last to keep the buildup on the tape down. You get sharper/crisper edges that way, too.
