at least on my bike you dont have to pull the carbs to set the pilot screws. I do have to remove the gas tank and seat but that is it.

The mixture screws that you are referencing differ from the actual pilot jets. Starter fluid will probably start it but then it will die. Something happened when you changed the jets as it ran OK before, right?

Since no one has mentioned this, I will. You may be getting fuel to the carbs, but it isn't getting to the cylinders if your plugs are coming out dry. How many turns out are your mixture screws?
